Production of endoglucanase by a novel strain Streptomyces sp.TKJ2 and its optimization under submerged fermentation

Anewly filamentous bacteriawas recovered fromTikjda forest soil (Algeria) for its high endoglucanase activity. The nucleotide sequence of the 16S rRNA gene (1454 pb) of Streptomyces sp. TKJ2 showed a close similarity (99%)with other Streptomyces 16S rRNAgenes. Endoglucanase production by Streptomyces sp. TKJ2 was optimized for varying culture conditions following one factor at time (OFAT) approach.An initialmediumpH6 and incubation temperature 30°C were found to be optimal for endoglucanase production, after 7 days of incubation. Studies on nutritional factors revealed that the highest endoglucanase production was obtained in medium made up of 10 g l-1carboxymethyl cellulose, 3 l-1g yeast extract, 1 g l-1NaCl, 2 g l-1 NH4Cl, and 0.3 g l-1MgSO4. Among various carbon sources tested, the actinomycete secreted high level of endoglucanase on some waste such as wheat bran and sugar can.
